Water-soluble polysaccharides from Pleurotus eryngii fruiting bodies, their activity and affinity for Toll-like receptor 2 and dectin-1

چکیده

The mushroom cell wall contains polysaccharides that can activate cells of the innate immune system through receptors such as Toll-like (TLR) and dectin-1. In present study, Pleurotus eryngii polysaccharide fractions containing a 3-O methylated mannogalactan (1?3)/(1?6)-?-d-glucans were isolated extensively characterized by 2D NMR methylation analysis. Traces (1?3)-?-d-glucan (1?2)-?-d-mannan also observed. Affinity for TLR2, TLR2-TLR6 dectin-1 using HEK-cells expressing relevant receptor genes was tested. PeWN, mannogalactan, inactive towards whereas fraction PeWB, more ?-glucan, activated heterodimer. Activation human ?-glucan correlated with amount in each fraction. Nitric oxide cytokine supernatant levels D2SC/1 dendritic stimulated P. interferon-? low to moderate. results indicate immunomodulatory activity water-soluble is modest.

Decreased Levels of Soluble Toll-Like Receptor 2 in Patients with Asthma

Background: Recently, reports have indicated a role for the membrane form of Toll-like Receptor 2 (TLR2) in asthma pathogenesis. In this study we examined soluble TLR2 levels in serum and sputum of asthmatic and healthy subjects. Methods: Serum and sputum samples were obtained from 33 asthmatic and 19 healthy subjects. The asthmatics were classified into four groups according to the Global I...

Collaborative Induction of Inflammatory Responses by Dectin-1 and Toll-like Receptor 2

Toll-like receptors (TLRs) mediate recognition of a wide range of microbial products including lipopolysaccharides, lipoproteins, flagellin, and bacterial DNA, and signaling through TLRs leads to the production of inflammatory mediators. In addition to TLRs, many other surface receptors have been proposed to participate in innate immunity and microbial recognition, and signaling through some of...

Correlation between Salivary Toll Like Receptor-2 Concentration and Early Childhood Caries

Background: Early childhood caries (ECC) is a common health problem in the developing countries. Basic knowledge about the etiology and pathogenesis of ECC plays an important role in its prevention. Objective: To determine the relationship between salivary TLR-2 concentration and early childhood caries formation Methods: Twenty-Eight children with ages ranging from 36 to 71 months (15 in ECC gr...

Toll Like Receptor 2 and 4 Expression in Peripheral Blood Mononuclear Cells of Multiple Sclerosis Patients

Background: Multiple sclerosis (MS) is a T cell mediated autoimmune disease with unknown etiology. Appropriate MS therapeutic strategies need thorough understanding of both disease etiology and pathogenesis mechanisms. Ligation of TLR-2 and TLR-4 stimulates the production of several cytokines leading to CNS autoimmunity and neurodegenerative diseases. Objective: To find a relationship between M...

Recognition of non-self-polysaccharides by C-type lectin receptors dectin-1 and dectin-2.

The discovery of several transmembrane receptors expressed by antigen presenting cells, including those that detect and interact with specific sugar moieties on the surface of microbes, has improved our understanding of how immunity against infection is generated. This knowledge, in turn, prompted us to review such interactions with emphasis on C-type lectin receptors and a focus on the roles o...
